Radioimmunoassay of Benzoylecgonine in Blood and Urine

A sensitive RIA for benzoylecgonine in body fluids is described. The label is radioiodinated p-hydroxybenzoylecgonine. The antiserum from an Emit-dau cocaine metabolite kit (Syva) is used at a 1:600 dilution. The standards are benzoylecgonine in buffer. PEG-6000 is used to separate the bound and free fractions. Cocaine, ecgonine, and methylecgonine have cross-reactivities of 2%, 1%, and 0.1%, respectively, compared with benzoylecgonine. Cinnamoylcocaine has negligible cross-reactivity. The detection limit (defined as the mean response of blank samples plus 3 standard deviations) is 20 ng/ml benzoylecgonine for both blood and urine. The assay is simple to perform, speedy, and extremely economical.
